PA Settings

Hello everyone!
I've got a question regarding PA settings when practicing in the studio. 
Does anyone ever mess with the highs, mids, and lows?
If so, does this make a big difference. 

I've been thinking about experimenting with different settings but I'm not sure when to do what.
Also, if there are any particular settings you use, please let me know what and why.
I'm trying to get a better understanding so that I can get better sound for my vocals live and in the practice studio.

You cannot have a "unique standard setting" as every voice have is formants and is harmonics.

So you may need to reduce mid 800hz for someone or increase mid 670hz for someonelse
Someone may have a hissing voice at 6000 hz someone else having poor highs.

Who is helping set the vocals in the live setting? Is this a sound person you know or are you setting them yourself? If it is someone you know, you can seek their opinion. In the practice studio, I have always like just a little reverb on vocals, just gives a little warmer/fuller effect. As for dialing in highs/mids/lows, each room you sing in is going to require a little tweaking to get what you like.
